|We have no idea why generic diazepam doesn't work properly. Usually differences between a generic and a brand-name drug relate to isomers, but sometimes there are specific chemical differences. For example, the position of a hydrogen on a tetrazole ring, which many biochemists will tell you just doesn't matter...
Most non-MP medications are going to interfere in some way with the actions of the immune system. Small drugs, like diazepam will occupy receptors competitively. Direct activities, such as modulating Ifn-gamma, will exacerbate potential problems.
We suggest brand-name Valium as a palliation as it is not tough for our members to wean, and it doesn't exacerbate IP.

The mechanism underlying BDZ-induced inhibition of
HIV-1 expression is unclear. It is well known that cellular
activation is a key requirement for HIV-1 replication in infected
cells, and transcription factors that participate in HIV-1
expression could be key therapeutic targets for antiviral agents.
Nuclear factor kappa B (NF-kB) is an important transcriptional
activator of inflammatory mediators, including certain
cytokines, chemokines, and adhesion molecules (30), and it is
well established that activation of this transcription factor is
necessary for self-perpetuated HIV-1 replication in mononuclear
phagocytes (1, 17, 26, 29, 33). Thus, we hypothesized that
the mechanism of diazepam’s anti-HIV-1 effect involved an
inhibition of NF-kB activation.Nuclear extracts (31) were obtained
from 5 3 106 chronically infected U1 cells which were
induced by high-density plating and treated with diazepam, as
well as the other prototypical classes of BDZs, to determine
the effect on NF-kB activation. Although equal amounts of
nuclear extract (2 mg) were added for different binding reactions,
a decrease in NF-kB binding was observed in the diazepam-
and RO5-4864-treated cultures, whereas clonazepam
treatment had little effect (Fig. 3A and B). This differential
effect of the three BDZs was consistent with their effects on
p24 Ag production in U1 cells, presented above
My knowledge,on this topic is limited, but did they just proved that diazepam is inhibitor of nuclear factor kappa b ? Same thing that Olmesartan does beside modulating VDR and other nuclear receptors. Connected to earlier study it would make sense that inhibition of nuclear factor kappa b activation would result in inhibition of interferon gamma.Could this be additional reason why it excarbates IP in Th1 patients?
Or maybe I am wrong inhibition of interferon gamma leads to inhibition of nuclear factor kappa b?

|I am only allowed generic and sometimes i experience the stronger herx co relation.
Living in the land of the lost. JUST OLMECIP 5 x a day.Sarcoidosis Dx.
Bone disintegration,lungs,joint/muscle/stomach pain, diarrhea,incontinence. Weakness on my left side sweats,fatigue,neuropathy,headaches,mood swings,cognitive diss.
